We plan to use calculated UDF on resource requests which are calculated on save. Is this possible? We know that the UDF for resource request are only created when the workflow job is executed and not when the resource request is created. Will this work with calculated UDFs?

Yes, the workflow job takes care of this. After the resource request UDF have been created, we trigger the recalculation so that calculated UDF will work also on resource requests.
